Dortmund were dealt a pre-match blow when top scorer Serhou Guirassy pulled a muscle during the warm-up, but the late addition of Julian Brandt to the starting line-up would prove inspired.
The first real chance fell to Kaishu Sano, who fired wide from distance for Mainz. The hosts grew in confidence and came agonisingly close to taking the lead in the 26th minute when Paul Nebel met a sharp cross from Armindo Sieb, but could only direct his sliding effort against the post.
Dortmund punished this miss ruthlessly just 48 seconds later. Brandt received a clever pass from Maximilian Beier on the right and delivered a perfect cut-back for Svensson to fire home from close range.
Mainz were still reeling when Dortmund struck again in the 40th minute. After winning possession near their own box, the visitors launched a lightning counter-attack. Adeyemi carried the ball forward at pace, exchanged passes with Brandt and calmly finished past Robin Zentner for his second goal in consecutive matches.
Mainz emerged with renewed determination after half-time and created a decent chance in the 56th minute when Andreas Hanche-Olsen headed a Nebel corner straight at Gregor Kobel.
Henriksen made a double substitution in the 63rd minute, introducing Stefan Bell and Arnaud Nordin, but any hopes of a comeback were dashed just four minutes later. Zentner rushed out of his area to challenge Adeyemi after a long ball from Kobel, mistimed his challenge, and was shown a straight red card.
With Mainz down to 10 men, Lasse Rieß came on in goal at the expense of Hanche-Olsen.
The Mainz sub had to be at his very best to stop Brandt adding a late third as Dortmund extended their unbeaten start to the season to stay hot on the heels of leaders Bayern Munich after five games.

Adeyemi was a constant menace to the Mainz defence with his searing pace and direct running.
His goal - a clinical finish after a brilliant one-two with Brandt - showcased his composure in front of goal, while his explosive burst forward won the red card that effectively ended Mainz's resistance.
Recording the highest top speed of any player (35.78 km/h), Adeyemi consistently stretched the opposition and was instrumental in Dortmund's fourth consecutive Bundesliga clean sheet.
Despite being the most pressured player on the pitch (17 times under pressure), the 22-year-old attacker thrived with the responsibility and thoroughly deserved his Player of the Match award.
